BIOC 203: INTEGRATED LABORATORY TECHNIQUES I

INSTRUCTIONS:

Attempt Question one & any other two.

SECTION A: Answer any 3 Questions.

1.(a) Differentiate a cation exchange resin from an anion exchange resin.

(b) A solution containing glycine,glutamic acid and lysine amino acids at pH 6.5 was loaded onto a column of carboxyl methyl cellulose. The column was eluted with a pH 6.5 buffer,with an increasing gradient of pH. Predict with explanation the elution pattern.(17 ½ marks)

2.Asprin,Caffeine,acetaminophen and phenacetin are compounds found in analgesic tablets. Explain how you would identify the compounds found in a new analgesic product for Asprin,Caffeine and acetaminophen using High performance liquid chromatography (HPLC).(17 ½ marks)

3.Your job after college is with a food industry in Nairobi.You will be required to choose a quick and inexpensive laboratory technique for fatty acids identification in various brands of cooking oil.

(a) Which laboratory technique are you going to use for the analysis? Why?

(b) Give the principles involved in (a) above.(17 ½ marks)

4.Explain the principles involved in the separation of protein by salt precipitation technique. Use serum protein in your explanation.(17 ½ marks)

7.(a) Calculate the Hydrogen ion concentration of Blood plasma, pH 7.42.(3 marks)

(c) Given the molar absorbance coefficient of tyrosine in water as 1420l mol-1 cm-1 at 275 nm.What is the concentration of tyrosine in a solution?(2 ½ marks)
