In biochemical analysis, many enzymes – catalyzed reactions can be monitored by
conversion of NAD(P) to NAD(P)H. This is done by measuring the absorbance

at two wavelengths (260 nm) where both reduced and oxidized forms absorb and
at 340 nm where only NAD(P)H absorbs.

Calculate the concentration of NAD(P) and NAD(P)H in a mixed solution from
the following data obtained using 1 cm cuvet given that the molar absorptivity at
260 nm = 1.05 x 10(4) for NAD(P)H and 1.84 x 10(4) for NAD(P). At 340 nm
only NAD(P)H absorbs with molar absorptivity 6.22 L/mol/cm. The mixed
solution gave absorbances of 1.32 and 0.382 at 260 nm respectively.
